Principal Research Scientist, lab director and biochemist with broad skills and experience relating to experimental design and implementation Extensive experience in molecular biology, protein expression, purification and characterization, and a strong background in bacterial and phage genetics In-depth experience with DNA manipulation including PCR, site-directed mutagenesis, plasmid construction, sequencing, TILLING, and Array CGH Numerous skills relating to protein expression and characterization: scale fermentation (up to 10 liters), protein expression in a variety of hosts and numerous purification techniques; enzymatic assays, kinetic analyses, electrophoresis and Western immunoblotting, In-Cell Westerns, ELISAs, fluorescence spectroscopy, Circular Dichroism, generation of artificial membranes and in vitro membrane protein reconstitution Assay development, optimization and high-throughput screening (HTS) via robotic implementation Execution of primary, secondary screens and deconvolution assays. Completed HTS campaigns against oncology targets (signal transduction kinases) Structure-activity relationships (SAR) and lead optimization experience in small molecule inhibitor drug discovery program DNA and RNA microarray projects (specifically Array Comparative Genomic Hybridization (CGH)) to associate drug resistance phenotypes with genetic aberrations Computer savvy with software training including ActivityBase, SARgen, and ClassPharmer Supervisory experience training coworkers and subordinates Excellent organizational skills and highly motivated Innovative, reliable, and eager to apply, enhance and broaden existing skill set
